How do you address false positives for the Y2H technology?
We flag technical false positives, artifacts and highly-connected proteins. Our internal database of over 10,200 Y2H screens covering 20+ years of experience provides an invaluble resource of PPI data and false positives, enabling us to analyze your results in a global context and guide you towards the most relevant interactions.
For Y2H-based screens using small molecules (YChemH, Y2HChem), a dependency assay is performed to identify the interactions that are strictly dependent on the presence of the molecule.
What is a Predicted Biological Score (PBS)?
PBS scores are a unique property of our ULTImate Y2H platform, where a prey target is assigned a confidence score and false positives are flagged, thus revealing the most relevant interactions.
The implementation of our PBS scoring system can be found in our earlier publications: Nature (Rain et al., Nature 2001) and Genome Research (Formstecher et al., Genome Research 2004) showing indeed that high-confidence interactions are correlated with real protein interactions in vivo.
How should I send my sample?
If you are sending us DNA samples, you can simply send them in tubes with Parafilm by normal ground mail. They don’t need to arrive quickly as DNA is stable.
If you are sending us RNA or protein samples, please use a large package filled with sufficient amount of dry ice and ship it by express mail. This type of material is very fragile and must be kept at least at -20°C. We recommend at the beginn of the week to make sure that we can immediately store the samples at the appropriate temperature.
If you are sending a small number of compounds, you can ship powders at room temperature by normal ground mail. If we need to receive a larger number compounds, we will require compounds in DMSO in 96 well plates on dry ice (Micronic or equivalent).
